identification and sequencing of seven mitochondrial trna genes in khorasan’s native chicken

چکیده
the aim of current study was to investigate the nucleotide sequence of seven trna genes in mitochondrial gene of khorasan’s native chicken and identification of possible mutations in it. for performing it, this part of the gene was cloned using the specific amplification initiators and cloned in the plasmid ptg19-t vector in dh5α strain of e-coli after the sampling and extracting the dna from whole blood. finally, the recombinant plasmid was extracted from a recombinant colony and the sequence of it was identified after the purification. the results of the nucleotide blast of this peer showed 100 percent similarity to the same nucleotide sequences of the gallus gallus domesticus with access number x52392. in addition to that, it was revealed that khorasan’s native chicken shows a similarity of 99 percent to the other chicken breeds in nucleotide sequences of these seven genes. also, no nucleotide mutation was observed while sequence comparison of the produced fragments to the similar sequence of gallus gallus. the order of trna genes in the sequence studied fragment was entirely identical to the mitochondrial genetic sequence of the gallus gallus and it was as follows: trnatrp, trnaala, trnaasn, trnacys, trnatyr, cox1 gene, trnaser, and trnaasp, and cox2 gene.

although there are studies on pragmatic assessment, to date, literature has been almost silent about native and non-native english raters’ criteria for the assessment of efl learners’ pragmatic performance. focusing on this topic, this study pursued four purposes. the first one was to find criteria for rating the speech acts of apology and refusal in l2 by native and non-native english teachers...
15 صفحه اولmutational analysis of mitochondrial trna genes in patients with asthma
background: mitochondria are autonomous cellular organelles that oversee a variety of functions such as metabolism, energy production, calcium buffering, and cell fate determination. most recently, mitochondrial dysfunction caused by mitochondrial mutations played important roles in the pathogenesis of asthma. however, the frequency of mitochondrial trna mutations in asthma is largely unknown. ...
متن کاملSequencing and Molecular Analysis of ATP 6 and ATP 8 of Mitochondrial Genome in Khorasanian Native Chickens
In order to perform breeding programs and improve production of native chickens, preserving genetic diversity in different areas of Iran is important due to the reduced available population. Genome sequencing is considered the most functional approach to determine the phylogeny relation between close populations. The aim of the present study was the evaluation of the phylogeny and genetic nucle...
متن کاملIdentification, Sequencing and Stability Evaluation of Eight Reference Genes in Saffron (Crocus sativus L.)
Saffron (Crocus sativus L.) is the most valuable and expensive spice in the world. The stigmas of saffron are the source of valuable apocarotenoids such as crocin, picrocrocin and safranal. transcriptomic and expression studies of genes are important steps in investigating of secondary metabolites in plants. One of the important prerequisites for such studies is the existence of reliable and st...
متن کاملCloning of yeast mitochondrial DNA in the Escherichia coli plasmid pBR322. Identification of tRNA genes.
Fragments of mitochondrial DNA from Saccharomyces cerevisiae were joined to the Escherichia coli plasmid pBR322 by the poly(dA):poly(dT) tailing method. A total of 347 transformants containing DNA inserts were isolated. The size of the inserted DNA ranges from 0.5 to 5 kilobases. To screen for mitochondrial tRNA genes, we isolated mitochondrial tRNAs, end-labeled them with 32P, and hybridized t...
متن کاملGenetic Characterization of Argas persicus From Iran by Sequencing of Mitochondrial Cytochrome Oxidase I (COX1) and 16s rRNA Genes
Background: Argas persicus has a great importance for health and veterinary, it can transmit many infectious agents such as Borrelia anserina (avian spirochetosis) and Aegyptianella pullorum. Distinguishing Argasidae due to close morphological relationship is difficult. OBJECTIVES: In the present study, we performed molecular analyses based on PCR and sequencing of Amplicon derived from 16S rRN...
